how is your final signal? comparison to what?

Final signal is great! The new ZEN probe has 8.5X times basline. The same probe with only a BHQ has 2.5x-3x baseline. With baseline subtracted the ZEN probe is only slightly better than the BHQ probe. See the above link for more information.

Well, it really shouldn't be different at all than the BHQ probe with baseline subtracted unless there is some inherit improved cleavage or decreased cleavage as a function of the ZEN chemistry..sounds like it just might not have much of an impact. How did you normalize the 'quantity' of actual oligo that you loaded into the reaction?
